Methods in molecular biology (Clifton, N.J.) 2012Gel-filtration chromatography, also known as size-exclusion chromatography (SEC) or gel-permeation chromatography, is a useful tool for structural and conformational analyses of intrinsically disordered proteins (IDPs). SEC can be utilized for the estimation of the hydrodynamic dimensions of a given IDP, for evaluation of the association state, for the analysis of IDP interactions with binding partners, and for the induced folding studies. It also can be used to physically separate IDP conformers based on their hydrodynamic dimensions, thus providing a unique possibility for the independent analysis of their physicochemical properties.
