Plasmodesmata (PD) play an important role in plant growth and development and defense. The permeability of PD is strictly regulated. Here, we describe an assay for measuring the permeability of PD in Arabidopsis thaliana leaves, which relies on tracing intercellular movement of green fluorescent protein (GFP) upon transient expression of the protein-encoding plasmid delivered by particle bombardment.
