DNA-encoded library synthesis is an evolved combinatorial synthesis, using a split-and-pool strategy in 96-well plates, libraries containing millions to billions of compounds can be prepared by a combination of chemical and enzymatic synthesis. Here we describe the analytics methods used in the library synthesis with one cycle of synthesis as an example. © 2022. Springer Science+Business Media, LLC, part of Springer Nature.
